Subject: 4-yr Postdoc in vibrant London Fitzrovia: cryo-ET of in-cell prion assemblies @ UCL
Applications are invited for a postdoctoral fellow to investigate how prion assemblies form and spread in cells and tissues (mouse model under development) using genetic code expansion, bio-orthogonal labelling and cutting-edge microscopy. The post is funded for 4 years. Apply by 24 September – link below.

About Us: UCL Institute of Prion Diseases, Faculty of Brain Sciences
We are one of the world’s leading research centres for the study of prions and their wider relevance to neurodegenerative disease (https://urldefense.com/v3/__http://www.prion.ucl.ac.uk__;!!Mih3wA!FN90r2pK8yXiO6nnP7KJII5_TUfKMCH-qVppWzkZB6xhshoLAvw34kRTZDkVveDlGYxqwnJMRKfLf-pTxdBF$ <https://urldefense.com/v3/__http://www.prion.ucl.ac.uk__;!!Mih3wA!FN90r2pK8yXiO6nnP7KJII5_TUfKMCH-qVppWzkZB6xhshoLAvw34kRTZDkVveDlGYxqwnJMRKfLf-pTxdBF$ >). The Institute is highly multidisciplinary with close integration of structural and molecular biology, genetics, cellular and animal models, small molecule and immunotherapeutics and clinical research. MRC and UCL have jointly invested £30 million in provision of a dedicated new building for the Institute in the central London’s vibrant and exciting Fitzrovia area, with excellent transport connections and local cultural and social amenities.

About the Role
The post-holder will be recruited to an MRC funded research programme led by Dr Szymon W. Manka. The aim of the project is to investigate molecular pathways of prion pathogenicity by tracking fluorescent prion assemblies in cells and tissues and elucidating their structures using cryo-correlative light and electron microscopy/tomography (cryo-CLEM/ET). The successful candidate should have experience in cryo-ET and sub-tomogram averaging. Experience in single molecule localisation microscopy will be a plus.

Context
Prion-like spread of proteopathic seeds is seen in numerous neurodegenerative conditions, including the most common Alzheimer's and Parkinson's diseases, which makes this research project widely relevant and impactful. The grant holder and colleagues from the MRC Prion Unit at UCL have recently made major contributions to characterising ex vivo prion assemblies at near-atomic resolution (https://urldefense.com/v3/__https://www.nature.com/articles/s41467-022-30457-7__;!!Mih3wA!FN90r2pK8yXiO6nnP7KJII5_TUfKMCH-qVppWzkZB6xhshoLAvw34kRTZDkVveDlGYxqwnJMRKfLfzciI0yA$ ; https://urldefense.com/v3/__https://www.nature.com/articles/s41589-022-01229-7__;!!Mih3wA!FN90r2pK8yXiO6nnP7KJII5_TUfKMCH-qVppWzkZB6xhshoLAvw34kRTZDkVveDlGYxqwnJMRKfLf6_W1A4D$ ; https://urldefense.com/v3/__https://link.springer.com/article/10.1007/s00441-022-03676-z__;!!Mih3wA!FN90r2pK8yXiO6nnP7KJII5_TUfKMCH-qVppWzkZB6xhshoLAvw34kRTZDkVveDlGYxqwnJMRKfLf8UdILRz$ ), but mechanisms of prion replication and spread are not well understood. The aim of the project is to identify the sites and molecular determinants of prion propagation in cellular and tissue contexts.

Cryo-EM sample preparation (Leica GP2), screening (FEI Talos 120C) and cryo-CLEM (Leica THUNDER Imager), as well as a 12-GPU cluster for data processing are available on site. Part of the work will be carried out in collaboration with Electron Bio-Imaging Centre (eBIC) at Diamond Light Source Ltd (Harwell Science and Innovation Campus, Oxfordshire, UK), providing access to focused ion beam (cryo-FIB) milling and state-of-the-art cryo-EM facilities. A Titan Krios microscope with Falcon 3 and K3 direct electron detectors and BioQuantum energy filter, and other advanced cryo-EM equipment will also be available at Birkbeck College London.

Salary
The post is graded as Grade 7 and will be offered on an initial salary of £39,508. Progression through the salary scale, which ranges from £39,508 - £47,355 is incremental.

Appointment Details
The post is available on a 4-year fixed term contract in the first instance. Deadline for applications is 24 September 2023. You can find more information and apply here: https://urldefense.com/v3/__https://www.ucl.ac.uk/work-at-ucl/search-ucl-jobs/details?jobId=14203&jobTitle=Post-Doctoral*Research*Fellow__;Kys!!Mih3wA!FN90r2pK8yXiO6nnP7KJII5_TUfKMCH-qVppWzkZB6xhshoLAvw34kRTZDkVveDlGYxqwnJMRKfLf25qcTo1$
