Hi Aninda,
Complementing the suggestions provided by Guillaume, recently it as published a review regarding the most critical parameters for sample preparation of membrane proteins for cryo-EM: [ https://urldefense.com/v3/__https://www.mdpi.com/1422-0067/24/19/14785*B47-ijms-24-14785__;Iw!!Mih3wA!BMiTj-FflnmlxCXmTEa6DadVq1VB77xp4lk_BMGEQy8fEFUZTA2rR-VDzbAz_w857TlZteJpiCtEXAjm1VkKEL5UFSMKt2iJKA5i1uI6lQ$ | https://urldefense.com/v3/__https://www.mdpi.com/1422-0067/24/19/14785*B47-ijms-24-14785__;Iw!!Mih3wA!BMiTj-FflnmlxCXmTEa6DadVq1VB77xp4lk_BMGEQy8fEFUZTA2rR-VDzbAz_w857TlZteJpiCtEXAjm1VkKEL5UFSMKt2iJKA5i1uI6lQ$ . ]
In this review, is reported that for some proteins under 100 kDa, nanodiscs could be very useful to overcome size-related limitations in the application of cryo-EM SPA for smaller proteins. Moreover, there is a report of a voltage-dependent chloride channel (VCNN1) with a total structure weight of 42 kDa (PDB ID 7EK2) reconstituted in nanodiscs at 2.7 Å resolution: [ https://urldefense.com/v3/__https://www.nature.com/articles/s41467-022-30292-w__;!!Mih3wA!BMiTj-FflnmlxCXmTEa6DadVq1VB77xp4lk_BMGEQy8fEFUZTA2rR-VDzbAz_w857TlZteJpiCtEXAjm1VkKEL5UFSMKt2iJKA4BE7zTJA$ | https://urldefense.com/v3/__https://www.nature.com/articles/s41467-022-30292-w__;!!Mih3wA!BMiTj-FflnmlxCXmTEa6DadVq1VB77xp4lk_BMGEQy8fEFUZTA2rR-VDzbAz_w857TlZteJpiCtEXAjm1VkKEL5UFSMKt2iJKA4BE7zTJA$ ]
I hope that this could help you.
Best regards,
Carlos.
