Hi Qiaohui,

Is there an image you could share? Your description suggests the methylcellulose possibly getting old, but could be other things.

Best wishes,

Vlad

Subject: [3dem] Needing expert advice on cryosectioning for Tokuyasu method)

Hi everyone,

I have been working on trying to master the technique of cryosectioning and immunogoldlabelling. My embedded sections in (mixture of 1% methycellulose and 1.15UA) on final image look good but there seems to many round blobs like bubbles that persist on/under my sections. Would appreciate if experts in this technique would be able to give some advice on how to rectify this. Thanks!

Kind regards to all,

Qiaohui
