[3dem] Leica EM GP

Hi

We own Leica KF-80 and Leica EM GP and recently tried both Gatan Cryoplunge
3 and Vitrobot Mark IV.

Each machine has its own pros and cons. (here I omit KF-80 because
it wouldn't be available)

Leica EM GP:
pros:
* liquid ethane is temperature controlled. You don't need to keep watching.
* Blot from only one side. Therefore, you can "concentrate" a sample on
grid by the filtering effect if the sample is larger than the hole size. <-
For us, this is important.
* (in our experience) good for thick specimens.
cons:
* difficult to make thinner  and consistent thickness of ice.
* (probably our machine specific) temperature control for the chamber is
not so good.

To Ed: we don't usually use the stereo microscope, but as Li pointed out it
would be helpful as the blotting force is very sensitive to the changes of
forceps positions and need very careful optimisation.

Vitrobot Mark IV:
pros:
* easier to make thinner and and consistent thickness (or gradient) ice.
cons:
* ethane is not temperature controlled.

Gatan:
pros:
* simple
cons:
* not fully controlled.
