Correlative light and electron cryo-microscopy (cryoCLEM) opens new avenues in structural biology by enabling resolving protein structures in situ within cells. To this end, cryoCLEM combines the advantages of light microscopy and cryoEM. Light microscopy allows for the identification and localization of cellular components using specific labelling with fluorescent dyes, while cryoEM provides superior resolution down to the sub-molecular scale. Recent years have seen significant improvements in the available cryoEM and light cryo-microscopy technologies, resulting in the rapid introduction of new instruments by manufacturers. In a project funded by the DFG, we will develop a modular, open-platform sample transfer station to streamline the movement of cryo-samples between different sample holders along a cryoCLEM workflow.
We are looking for a motivated researcher who will be in charge of developing and overseeing the testing of the novel cryoCLEM sample transfer station.

TPLM consists of several light microscopy facilities, including the Advanced Light and Fluorescence Microscopy (ALFM) facility at the Center for Structural Systems Biology (CSSB) Hamburg (https://urldefense.com/v3/__https://www.cssb-hamburg.de__;!!Mih3wA!FFIWCKanMUUp1W9Aps1zlecbkgtWwEsklXIKzQtYMul2MnSZF4YJjH-U8vVoJcxZx6cbP1fNV54aw9ANiy5ylfJVA26gvw$ ). The ALFM facility has experience in developing tools for cryoCLEM workflows and closely collaborates with the cryoEM facility at CSSB.
The recruited candidate will be supported by the ALFM facility team during the whole project. Furthermore, the cryoEM facility team and CSSB researchers will provide feedback to achieve a well-usable design of the open-platform cryoCLEM sample transfer station. The recruited candidate will also be responsible for documenting and publishing the designs and testing results of the open-platform cryoCLEM sample transfer station, as well as supporting researchers in using the transfer station.
Role requirements:
* thorough understanding of the principles of correlative light and electron cryo-microscopy (cryoCLEM) and its application for biological research
* profound expertise and experience in developing and implementing novel research methods
* knowledge and experience in designing and building tools or components for light or electron microscopy, preferably for cryo-microscopy
* experience with using CAD design software (e.g. Autodesk Inventor, SolidWorks)
* experience in working with light-cryo-microscopes or cryoEMs is an advantage
* proficiency in English (spoken and written, B2 or better)
* initiative, planning and organizational skills
* very good communication, ability to work in a team and resilience
