Postdoctoral Training Fellow: Cryo-Electron Microscopy and Biophysics/Biochemistry

Following a Cancer Research UK Programme Foundation Award to Dr Sebastian Guettler, a Postdoctoral Training Fellow position is available in the Structural Biology of Cell Signalling team in the Divisions of Structural Biology and Cancer Biology. Work in the Guettler laboratory centres on the elucidation of signal transduction mechanisms relevant to the function of cancer cells, in particular the Wnt/beta-catenin network and telomere homeostasis and their control by poly(ADP-ribosyl)ation. Research by the team combines structural biology, both by X-ray crystallography and cryo-electron microscopy, biochemistry, biophysics and functional studies in mammalian cells. The Cancer Research UK-funded Postdoctoral Training Fellow will analyse the function of poly(ADP-ribose)polymerase (PARP) enzymes by using a combination of cryo-electron microscopy, biophysical and biochemical approaches. The emphasis will be on the structural characterisation of proteins/protein complexes by cryo-EM and the exploration of hypotheses raised by structural findings using purified proteins in biochemical assays. The position will coincide with a major expansion of cryo-EM capabilities at the ICR in 2019/20, which will provide a state-of-the-art infrastructure and outstanding research and training opportunities.

Research in the Division of Structural Biology is aimed at understanding the structural basis for the function and regulation of proteins and protein complexes implicated in cancer. Our research programmes are complemented by close collaborations with other groups at the Institute, for example in the Divisions of Cancer Biology and Cancer Therapeutics. The Division of Structural Biology has managed facilities for protein production, with expertise in multi-subunit expression (insect cell, yeast and bacterial expression, including a 60-litre fermentor), protein crystallography and cryo-EM (FEI Tecnai T12 and F20). The installation of a state-of-the-art FEI Glacios/Falcon 3 cryo-electron microscope is currently in progress. Regular access to shared cryo-EM facilities, such as eBIC, is available. Moreover, the ICR is part of the London Consortium for Cryo-EM, hosting an FEI Titan Krios equipped with a Volta phase plate, a Gatan Bioquantum energy filter and K3 detector (available in 2019). The Division is also well-equipped with instrumentation for biophysical analysis (e.g., ITC, fluorescence, multi-angle light scattering).
