Hi Chuang,

We use SerialEM for this, it allows to control frames for each imaging mode so you don’t have to switch frames on and off constantly, something FEI software can not do. Also see Chen Xu’s (Brandeis) fantastic manuals on how to do "EPU-style" acquisition with SerialEM for only careful selected targets: http://www.bio.brandeis.edu/KeckWeb/emdoc/en_US.ISO8859-1/articles/

But for our filament project, EPU haven't give us such overwhelming effect as in single particle field. In our situation, the filament's concentration is kept low to avoid overlapping,so the chance to shoot a good tube is not high when automatically collecting data hole by hole. We still prefer manual data collection by taking an image at low mag(4300x) and moving stage to the target area for higher mag imaging. And here it seems that this method is more efficient.

So does anybody have experiences on such situation that either collecting data manually with Falcon's movie mode or collecting data while running EPU but under supervision?
