[3dem] gold fiducials

Hi Oleksiy,
We do carry 10nm or other size gold covered with PEG. It does not contain “peptidol”, but it disperses exceptionally well and has no surface charge. It is not on our website yet, so contact me off list if interested.
In my days doing tomography, I did try BSA-gold, Protein-A gold, even IgG-gold, besides unconjugated colloids. Protein-stabilized gold worked very well, unless there was an interaction like described in the OP.
Best wishes,
Vlad

Hi Sharon,
is this PEG-nanogold available for purchase somewhere?
Thank you,
Oleksiy

Hi Daniel,
I can confirm that these gold fiducials produced by Laurence Duchesne behave very nicely. We use them routinely here.
They do not usually stick to macromolecular or cell preps.
Best,
Sharon

Dear Daniel,
We routinely use 10 nm gold NPs coated with a mixed PEG-peptidol matrix prepared according to :
Robust ligand shells for biological applications of gold nanoparticles

We rarely see interactions with proteins. They occasionally tend to stick to some types of lipid vesicles, notably at the air-water interfaces.
Best wishes,
Denis

Hi Daniel,
We use these…
BSA-treated tracers, EM grade (EMS, cat. no. 25486).
You can also make your own BSA-treated gold, protocols outlined in Iancu et al., Nature Protocols (2006).
Good luck!

Dear All,
The 10 nm ProteinA gold fiducials that we use in cryo-ET experiments strongly interact with one of our proteins of interest. Do you have any suggestions for a 10 or 6nm gold fiducials ?
Best
Daniel Lévy
