scientists seeking to match their scientific passions with an appreciation
for the great outdoors.
We are seeking scientists at all levels that are interested in
understanding the mechanisms by which bacteria acquire immunity to invading
parasites, like viruses and plasmids. Acquired immunity in prokaryotes is
achieved by integrating short fragments of foreign nucleic acids into
clustered regularly interspaced short palindromic repeats (CRISPRs). These
repetitive loci serve as molecular ‘vaccination cards’ by maintaining a
genetic record of prior encounters with foreign transgressors. The CRISPR
locus is transcribed and the long primary transcript is endonucleolytically
processed into a library of short CRISPR-derived RNAs (crRNAs) that each
contain a unique sequence complementary to a foreign nucleic acid
challenger. Each crRNA is packaged into a large, multi-subunit surveillance
complex that patrols the intracellular environment and ‘silences’ invading
foreign nucleic acid targets. Work in our lab uses a wide range of
genetic, biochemical and biophysical approaches to understanding how these
surveillance systems efficiently find and bind foreign DNA on a time scale
that outpaces viral replication.
Position will be available starting in June (2012) and funding is available
