A postdoctoral fellow is needed for structural analysis of the eukaryotyic multienzyme complex of aminoacyl-tRNA synthetases. The project focus is to test and refine the three domain model of the multisynthetase complex by calculation of its three-dimensional structure from cryoelectron microscopic images and by mapping locations of its constituent polypeptides using labeled tRNAs and antibodies as structural probes.
A starting date of April - May 2001 is ideal, but later dates will be considered, if necessary. The position will be funded for at least 3 years by NSF.
Required experience:
Ph.D. in Biochemistry/Biophysics/Biology with specialization in computational microscopy.
Specific skills must include: acquisition of cryoelectron microscopic data and calculation of three-dimensional volumes using single particle methods in SGI IRIX environment (SPIDER/WEB and EMAN packages are currently available)
Other desired skills: basic biochemical methodology including protein and nucleic acid purification and labeling, HPLC operation
Pertinent equipment:
Three year old LEO 912AB energy filtered microscope
Oxford cryoholder and LEO cryobox for preparing specimens
SGI Octane computers with connection to 64 processor SGI Origin
