<p>these are some things I still remember from my Eindhoven days and
that you might want to consider (all are my personal opinion):<br>

<p>1. throughput: if the Autoloader (AL) is warm, cassette
docking/sample loading will take longer than in cold state (as it
takes longer for AL vacuum to recover). This will worsen with
increasing number of docking rounds. Sample load = coffee break at
some point.<br>
</p>
<p>2. If your customer wants to image multiple samples in one day
(i.e. many sample exchanges) you might want to <i>consider asking
your Service Engineer</i><i> to</i> temporarily change/loosen
the AL (PP AL) vacuum trip values to speed things up (trade-off of
vacuum quality and speed).<br>
</p>
<p>3. You'll need to schedule a decent round of Autoloader and
Column conditioning after a room temp session (especially if you
have decided to go for 2.). That adds to the time needed for this
session.� </p>
<p>4. I think it's the (mechanically) safe option to operate
Autoloader and column both at room temp - I'd never dare to
operate column warm & AL cold.� I'm not even sure if all Talos
SW versions allow sample loading with "one cold one warm"??</p>
<p>5. With conditioning column and Autoloader after a room temp
session, there is no reason to assume the column is "dirty" or
"contaminated" by any means.</p>

for single-particle cryo-EM and cryo tomography.<br>
