motivated and
creative Post Doctoral Researchers for structural and molecular
biology studies
to </span><span

how DNA
replication is initiated and regulated in the cell cycle</span><span
style="font-family:"Arial","sans-serif";mso-fareast-font-family:"Times
New Roman";
mso-fareast-language:EN-GB">. The research will be performed by
the team led by
Prof Elena Orlova (ISMB, Birkbeck, London) and Dr. Cyril Sanders
(</span><span
style="font-family:"Arial","sans-serif"">University
of Sheffield). In the lab
by Dr Sanders the discrete protein-DNA complexes will be
assembled, purified
and characterised for their distortions, melting and then
unwinding of the DNA
double helix. Structures of replication complexes will be
determined using cryo
electron microscopy (cryo-EM) in the lab of Prof Orlova at
Birkbeck College
(ISMB) in London. The opportunities offered by the “resolution
revolution” in
cryo-EM make this is an exciting time to be involved in
mechanistic
biochemistry. The full-time posts are funded by the
Biotechnology and
Biological Sciences Research Council (BBSRC) and are available
for a period of

for cryo-EM
analysis of </span><span
style="font-family:"Arial","sans-serif";color:black">complexes
formed by two viral proteins, E1 and E2 and a specific dsDNA
sequence to
understand replication initiation. Cryo-EM and single-particle
analysis (SPA)
allow visualisation of particles in solution under near-native
conditions. The
specific objectives are to determine the structure of the
E1E2-ori
pre-initiation complex and reveal the ATP-dependent
conformational changes in
the protein and DNA, to derive a high-resolution cryo-EM
structure of the E1
helicase bound to a replication fork junction (RFJ)-like DNA
substrate. It is
important for both experimental design and interpretation of
results to have
active scientific interactions with the collaborating
laboratories (Dr. C.
Sanders, University of Sheffield).</span><span
style="font-family:"Arial","sans-serif";

applicant will interact with members of the Birkbeck Electron
Microscopy group (<a

equivalent) and
be accomplished in biochemistry and structural biology<span

layout-grid-mode:line"> or a related area, and have relevant
research
publications.</span> Expertise in cryo-EM structure
determination would be
especially desirable. </span></p>

laboratory is equipped with a high end Polara microscope and
three other Tecnai
microscopes including a 200 kV FEG system. Our Polara has a K2
direct electron
detector and F20 has a DE camera s that provide high resolution
data. Funding
has been obtained to purchase in Titan Krios with direct
electron detector. The
EM lab has the associated facilities for sample preparation,
including a
vitrobot, and a dedicated linux cluster and graphics
workstations for image
processing. The Birkbeck EM lab is a major focus for EM research
and training:
we host an EMBO course on cryo EM single particle analysis every
2 years, and
also an increasing number of successful research fellows. </span></p>
