Workshop on Image Processing with 2dx – August 19-25, 2012, in Basel, Switzerland.

We invite to the fourth international workshop on electron crystallography of membrane proteins. This workshop will focus on data processing, with emphasis on 3D reconstructions from images of tilted 2D crystals, using the 2dx software package. 

This hand-on workshop on the processing of images from cryo-electron microscopy of 2D crystals of membrane proteins (electron crystallography) will provide the theoretical background and practical applications in advanced image processing algorithms. The workshop will use the 2dx software system, which is based on the MRC programs.  Theoretical morning lectures will be followed by practical exercises in the afternoon. Although we will provide computers (iMacs), you are encouraged to bring your own laptop (Linux or OSX), or at least a portable hard-drive (USB2.0), onto which we will install the relevant software systems and test data during the workshop.  You are also welcome to bring your own dataset. 

The 2dx software system offers a graphical user interface to run the MRC programs in a transparent, user-friendly, and efficient way. Additional programs complement the MRC package, among others to automate the processing.   

The recent version of 2dx allows fully automated processing of images of non-tilted and of tilted 2D crystals. The 2dx_merge module allows user-friendly analysis of the data, determination of the common phase origin, 2D and 3D merging, scaling of amplitudes with respect to a reference dataset (SCALIMAMP3D), lattice line interpolation (LATLINE), refinement of the defocus, tilt geometry and beam tilt from the 3D dataset (ORIGTILT), 3D calculation of a final volume, sub-volumes, and point spread function, and automated re-processing of the entire image data using the 3D structure as a reference (MAKETRAN), followed by re-merging. Difference map calculation in Fourier space is supported. A maximum likelihood based single-particle module allows structure reconstructions also from less-well ordered crystals.

We explicitly also invite experienced users of the MRC software to join this workshop, to foster discussions and experience exchange on the optimal usage of those routines.
