Bill Baxter wrote Matlab code for reading Spider files:
Read SPIDER files - File Exchange - MATLAB Central

For other EM formats I have code on the Matlab File Exchange too.
Imagic, MRC and DM3 file i/o - File Exchange - MATLAB Central
