I got my Ph.D degree in crystal structure analysis of
some anti-HIV active nucleoside analogs. Soon after
submission of my Ph.D. thesis I worked on RecX (a RecA
regulatory protein of mol wt 19kDa), its expression,
purification and crystallisation. I went to USA to do
my postdoctoral research work on blood coagulation
protein Prothrombin (72kDa), its crystallisation and
X-ray diffraction analysis of factor VIIa-TissueFactor
complex using programs X-PLOR and O. At present in
India I am doing collaboration work on crystal
structure analysis of cysteine protease Calotropin
(29kDa) by molecular replacement using programs
X-PLOR and O.
I am very much interested in your research work. As I
have got research experience in protein
crystallography right from expression, purification
and crystallization and crystal structure analysis so
I find that my expertise fits your laboratory. I have
great respect for you and your research work. I think

Experience: Research experience in molecular biology
in studying structure-function relationships of
drugs,proteins and enzymes by X-ray diffraction and
physicochemical methods. I got my Ph.D. degree under

Research for Ph.D. degree :
My Ph.D. thesis was on crystal structure analysis of
some anti-HIV active nucleoside analogs by X-ray
diffraction to study molecular mechanism of drug
action at the protein and gene level.

In India after submission of my Ph.D. thesis I worked
on expression, purification and crystallisation of
RecX (19KDa) regulatory protein of RecA under Dr.Amit
Das in the department of Biotechnology of Indian
Institute of Technology, Kharagpur, India.
Postdoctoral research in USA:
Period October 2001- September 2002
In USA I did my postdoctoral research work on some
blood coagulation proteins Prothrombin (MW 72000) and
FVIIa/TF complex in order to crystallise and determine
structure by X-ray diffraction in the department of
Hematology and Oncology in Saint Louis University. I
screened a wide range of crystallisation conditions
for crystallisation of prothrombin with inhibitor
complex using the sparse-matrix method and got
crystallising conditions and crystals. I also worked
on 2.8Å resolution data for FVIIa/TF complex in order
to refine the structure using programs X-PLOR and
O.
Continuation of research in Protein Crystallography:
Period October 2002- in India
In India at present I am doing collaboration work with
Prof.Ashoke Banerjee,Head of the Department of
Biophysics,Bose Institute,Calcutta on crystal
structure analysis of cysteine protease
calotropin(mol. wt 29kDa) by molecular replacement
using the program X-PLOR and interpreting the electron
density map using the program O.
Expertise :
A) Crystallisation and Crystal structure analysis of
small molecules by X-ray diffraction methods.
B) Crystal structure analysis of protein molecules by
X-ray diffraction methods.
C) Protein expression, purification and
crystallisation by dialysis and vapour
diffusion(hanging/sitting drop method).
D) Basic biochemical characterisation (quality
control) of the protein samples to crystallise.
E) Optimisation of crystallising conditions to produce
crystals suitable for X-ray diffraction.
F) Experience in the use of softwares like X-PLOR,CNS
and O.
